Due to ETC's new non-fluorescence gel-support the fluorescent staining methods getting possible! After 2 hours reaction with a FITC-chromogen conjugated antibody (DAKO F0202) and 3 hours washing out of the exceed antibody, the gel goes directly into a Fluorescence-Imager. The FITC-antibody's physical properties: 495 nm excitation, 522 nm emission.
Advantage: Quick, easy, sensitive and IgG-selective staining. Disadvantage: Additional hardware is necessary.
Tested instruments:
GE Healthcare's Typhoon series: Settings of the Typhoon-Imager (GE-Healthcare): Blue laser, 488 nm excitation, resolution: 40 µm, amplification: 450 PMT-voltage, filter: 522 nm, focus (platen):0.
Fujifilm: FLA 7000 and FLA 5100: Settings: Blue laser, 40 µm resolution, amplification 825 pmt, filter 530 DS “FITC”
